Could anyone explain How is this procedure?
Cryopreservation in liquid nitrogen (-190 oC) helps spermatozoa to keep their activity for a long time enabling the use of sperm in various assisted reproduction techniques (IUI, IVF, ICSI). The tremendous developments in the field of assisted reproduction have helped a lot of men suffering from cancer or other diseases which could lead to infertility to finally have children of their own. A careful cryopreserved sperm or testicular tissue remains vital for an indefinite period of time (20 years) after which they can be thawed and used for IUI or other methods. Patients who will decide to cryopreserve their sperm should abstain from any sexual activity for at least 2 – 5 days before giving a sample and they should have completed screening for HIV and Hepatitis B, C and VDRL at least 30 days prior to the day they will give the sample.

Cryopreservation. Dead Or Alive?

Are humans who volunteer to be cryopreserved, frozen while they are still alive? The whole thing makes absolutely no sense. (Please spare explanations on the whole process. No cheek intended but I know how it's done. I just fail to see how preserving someone after they've died can possibly mean a chance for regeneration or awakening, much less done if they were alive. I do realize that sperm cells, embryos and ovarial things have been preserved successfully.)
People who are cryogenically frozen are dead when the process is begun. Usually they have died of cancer, AIDS, or another heretofore incurable disease/disorder, and hope to be 'brought back to life' when the cure is found. The theories behind cryogenics are sound, but we have yet to find a method to thaw out and then reanimate the body. The cells are destroyed during the freezing process, and so far, no one except Frankenstein knows how to bring dead tissue back to life.

Objective: To evaluate the effects of cryopreservation on sperm mitochondrial activity and nuclear DNA integrity in men with spinal cord injury.Design: Prospective controlled study.Setting: Patients in an academic research environment.Patient(s): Men with and without spinal cord injury-induced anejaculation.Intervention(s): Electroejaculation or penile vibrating stimulation semen cryopreservation using a commercial TEST-yolk-buffer technique.Main Outcome Measure(s): Rate of sperm DNA fragmentation as assessed by the comet assay, graded in Classes I (high DNA integrity) to IV (high DNA fragmentation). Mitochondrial activity as assessed by a method in which active mitochondria precipitate 3,3?-diaminobenzidine. Cells were classified as I (all active) to IV (all inactive). Semen was cryopreserved in a Test-yolk buffer, and motility, DNA fragmentation, and mitochondrial activity were analyzed precryopreservation and postthaw.Result(s): Before cryopreservation, when the study (SCI) and control groups were compared, no statistically significant differences were found with respect to concentration or total sperm count; however, the SCI group presented significantly lower ejaculate volume, decreased sperm morphology, and an increase in the round cell and neutrophils counts. In both groups, cryopreservation was associated with an increase in DNA fragmentation, a decrease in mitochondrial activity, and a decrease in motility, of which the latter was of greater importance in the control group.Conclusion(s): Cryopreservation causes a decrease in conventional seminal variables as well as in mitochondrial activity and DNA fragmentation. However, these were no more detrimental to sperm from men with SCI than to sperm from the control group.
